They do not use shards, nymphs, marks, relic, or (for some reason) miracle. Anything else (upped or unupped) is possible IIRC.

edit: In case of pedantry, you also can't win malignant cell, ash, shard golem, or any non-card card I'm forgetting from them.

Apart from shards and non-obtainable cards (Malignant cell, ash, shard golem..) you can gain literally anything from them, as their deck is randomized.

are the chances of receiving upped cards from AI4 more than AI3? If the chance is greater is it worth the time investment for the longer games?

There is no chance of winning upped cards from AI3. Which one is better to farm has been discussed here.
