Just curious, if you were upgrading this from scratch. Would you go towers, graboids, Shriekers or Antlions first, second and so forth?

Normally I recommend towers first, but antions are terrible unupped.  I would probably do them first or atleast very early.

I don't have antlions yet, I actually started with the unupped Shrieker deck in your Turns to win study.

In that case, up 14 towers 1st, then add antlions 1 by 1 as you up them, removing pillars and shriekers as you go, up the graboids, sword, and finally shriekers.
