According to the new Notes, creatures are not considered "ready" unless they glow.
Yes, you can "save" Mind Flayer from being delayed by taking away its readiness before playing Rotation.
Quote
"Ready" means unaffected by summoning or activation sickness. It is displayed by the glow on creatures who can activate their abilities.
This does not communicate that the glow is required for it to be considered "Ready"

So Rotation is:
Use all the activated abilities of your Readied creatures
[optional] Play Mitosis/Butterfly Effect or use Mutation on your opponent's creatures
Play Rotation
Use all the activated abilities of your creatures
The creatures with activated abilities your opponent controls are delayed for 2 turns

No, you need to play 2 Rotations in one turn to delay your opponent's creatures.
As implied by the notes, the creatures without activated abilities that your opponent controls will also be delayed.

A creature is "ready" iff they are glowing.
If a creature is not glowing, they are not "ready" and are affected by the first mechanic of Rotation as expanded in the Notes.
If a creature is not affected by summoning sickness but has no ability as of yet, it is not glowing, and therefore is not "ready", because its "readiness" is not displayed, as explained in the Notes.

In this case using a creatures ability and then playing this card would define the creature as "not ready" and I may be mistaken, but you never see a "glow" around the opponent's creatures.

Using a creature's ability and then using this card would cause the creature to glow and "define" the creature as ready.
True, there is no way to see glowing creatures on the opponent's side of the field in the current version of the game. That does not mean it's impossible to make a card that causes the creatures on the opponent's side of the field to glow.
