So only after the debuff has been executed you can choose a target for the buff.  - Opponents creatures are prefered (twice as often) for the debuff. Update 1: Text fixed.

Are the effects sequential or simultaneous?

Sequential.
So if this is the only creature on the field and you use its ability, the Xeno. Spirit dies (it gets -2/-2 or -2/-3 upped) and you can choose a target (though you won't fine any it there isn't a creature left). It is comparable to Virus.

if you want to be precise, use 'up to' clause in the text

i.e.
